Introduction

The “Uncle Ben’s Tek” is a simple method for learning sterile technique and grain-to-bulk transfers. It uses pre-cooked rice bags as a starter medium and can be adapted for educational or experimental purposes.


Materials Needed

  • Pre-cooked rice bags (Uncle Ben’s or equivalent, 90-second microwave style, unopened)

  • Sterile syringe with solution (spore, liquid culture, or other inoculant)

  • Alcohol wipes or 70% isopropyl alcohol

  • Flame source (lighter or alcohol lamp)

  • Micropore tape

  • Scalpel or small sterile blade (optional)

  • Sterile gloves and face mask

  • Clean working area or still air box

For Spawn to Bulk:

  • Sterilized bulk substrate (coco coir, vermiculite, or similar)

  • Monotub, shoebox, or container with lid

  • Liner (optional)

  • Spray bottle with clean water


Step 1: Preparation

  1. Clean your work area thoroughly with isopropyl alcohol.

  2. Wash hands and wear sterile gloves.

  3. Shake rice bag gently to break up clumps.


Step 2: Inoculation

  1. Flame sterilize the needle until red hot. Let it cool for a few seconds.

  2. Wipe the injection port area (corner of bag) with an alcohol wipe.

  3. Insert the needle through the bag (just above the rice) and inject 1–2 mL of solution.

  4. Seal the hole with micropore tape.

  5. Gently massage the rice to distribute solution.


Step 3: Colonization

  1. Store bags in a clean, dark area at 70–78°F (21–25°C).

  2. Within 1–3 weeks, you should see white growth spreading from the injection point.

  3. Once the rice is fully colonized, wait an extra 5–7 days to ensure consolidation.


Step 4: Spawn to Bulk

  1. Prepare your container (monotub/shoebox). Optionally line the bottom with a plastic liner.

  2. Break up the colonized rice bag gently into small pieces.

  3. Mix with pre-hydrated bulk substrate at a ratio of about 1:2 (spawn:substrate).

    • Example: 1 bag of rice to 2 bags of coco coir mix.

  4. Level the surface but do not pack it down tightly.

  5. Cover with lid. Some prefer to place a thin “casing layer” of substrate on top.


Step 5: Incubation

  1. Place the container back in the clean storage area.

  2. Allow the mixture to colonize for 1–2 weeks until the surface is fully white.


Step 6: Fruiting Conditions

  1. Once fully colonized, introduce fresh air exchange by loosening the lid or adjusting tub holes.

  2. Mist the sides of the container lightly if surface looks dry.

  3. Provide indirect light on a 12/12 cycle.


Troubleshooting

  • Green, black, or pink spots → contamination; discard bag safely.

  • Slow growth → temperature may be too low.

  • Overly wet rice → may stall; use less inoculant next time.

  • Dispose of contaminated materials responsibly (sealed trash bag).
